MMP3 και MCP1 στον πνεύμονα σε μοντέλο Ι/Ε ήπατος με χορήγηση απιγενίνης (Master thesis)

Introduction. In liver and transplant surgery, it is often necessary to perform ischemia-reperfusion in order to treat the main disease of the patient. The damage, however, from this surgical time, affects not only the liver but also other remote organs, such as the lung. Thanks to the grade advancements in medical science, along with the valuable aid of technology and studies from experimental protocols, constantly improvement has been evident both in morality rate as well as in post - operative complications. There is a grade likehood that the administration of antioxidant substances, such as apigenin, acts protectively after ischemia-reperfusion in the liver parenchyma. The results are documented by the expression of MMP-3 and MCP-1. Aim. The aim of the present experimental protocol, which was conductive on rats, was studie and investigation of the protective effect of the antioxidant substance apigenin on the lung after ischemia-reperfusion in the liver parenchyma. The evaluation of the results was made by measuring the tissue expression of MMP-3 and MCP-1. Materials and Method. Sixty-three male albino Wistar rats, approximately 15 weeks old and weighing between 220 and 350 grams, participated in the study. The rats were randomly divided into three groups. The first group included those who underwent a sham operation (Sham group), the second group the rats who underwent ischemia-reperfusion but were not given apigenin (group C - control), and the third group the rats who underwent ischemia-reperfusion and were given apigenin intraperitoneally (Ap – apigenin group). The C - control and Ap - apigenin groups were divided into four equal groups based on reperfusion times at certain time intervals, such as 60 minutes, 120 minutes, 180 minutes and 240 minutes.Results. The change in the degree of variation of both MMP-3 and MCP-1 is particularly evident and much smaller in the groups of rats given apigenin at fixed time intervals. In contrast, in the groups of rats that underwent only ischemia-reperfusion at the same times, there is evidence of a comparatively greater tissue expression of the markers. As for the group of rats that only underwent the sham operation, no changes in the levels of MMP -3 and MCP -1 were observed. Conclusions. The protective role of the administered antioxidant apigenin during liver ischemia-reperfusion is confirmed by the expression of MMP-3 and MCP-1 in the rat lung. The results of the present provide concrete for optimism indication in the particularly difficult and demanding field of the liver surgery.
