Μελέτη του υποδοχέα της ιντερλευκίνης-6 (IL-6) σε ασθενείς με Μυελοδυσπλαστικό Σύνδρομο υπό θεραπεία με Αζακιτιδίνη (Bachelor thesis)

Myelodysplastic syndromes (MDS) are a heterogeneous group of clonal hematopoietic stem cells disorders characterized by ineffective hematopoietic capacity and high probability of evolving into acute myelogenous leukemia (AML). In patients suffering from high-risk MDS, the administration of hypomethylating agents is recommended as the mainstay treatment. Of course, their mechanism of action has not been fully elucidated, although recent research has shown that they activate antineoplasmatic immune response. In the present study, the correlation between IL6 / STAT3 activation levels in patients with MDS and the response to treatment with azacitidine hypomethylation agent was evaluated. To reach the above mentioned goal, a mononuclear leukocyte layer was isolated from peripheral blood samples in pretreatment patients and during the 6th cycle of azacitidine treatment. These cells were incubated with the respective stimulants and the results were analyzed by phospho-specific flow cytometry. The experiment results showed that the patients with low STAT3 transcript levels after IL6 stimulation had a higher average overall survival. It was concluded that the IL6 levels and its receptor are not correlated with the response of the patients who are treated with azacitidine.
