Μελέτη της διαφορικής έκφρασης των κινασών ΑΚΤ1 & ΑΚΤ2 στο στρώμα και στο επιθήλιο των μαστικών αδένων ποντικού και των επιδράσεών της στην πολλαπλασιαστική ικανότητα των επιθηλιακών κυττάρων (Bachelor thesis)

The development of the mammary glands is regulated by the action of hormones, growth factors as well as intercellular interactions which lead to the activation of intracellular signaling pathways. A key signaling molecule with a critical contribution to both normal growth and mammary gland oncogenesis is the family of AKT kinases. The present thesis investigates the correlation of the differential expression of AKT1 and AKT2 kinases in the mouse mammary gland stroma and epithelium with their ability to regulate the development and proliferation of mammary epithelium during gestation. For this purpose, a study was performed on the proliferative activity of mammary gland epithelial cells in three categories of genetically modified mice: wild type (AKT+/+), knock out of AKT1 and AKT2 isoforms (AKT1-/- & AKT2-/-) and chimeric tissues composed of: a. AKT1-/- epithelium and wild-type stroma and b. AKT2-/- epithelium and wild-type stroma. The developmental stage chosen to be studied was day 17.5 of pregnancy (p17.5) in which cell proliferation is observed and the method used to conduct the study was the immunohistochemistry technique. The results showed that the effect of AKT1 kinase on the proliferation of epithelial cells is cell autonomous and depends only on its expression in the epithelium while the effect of AKT2 on the proliferation of epithelial cells is stroma-dependent. This is evidenced by the fact that complete loss of expression of AKT1 kinase in the mammary gland and loss of its expression only in the epithelium but not in the stroma, show the same phenotype, while, respectively, complete and partial loss of expression of AKT2 kinase show a different phenotype.
