Non-small cell lung cancer (NSCLC) has been implicated in 80-90% of lung cancer cases worldwide, accounting for a low five-year overall survival in both men and women. The great efficiency of radiation in killing tumor cells, has established radiotherapy as the leading cure for patient with NSCLC. Recent research on immunological effects of RT suggests that radiation-induced DNA damage triggers tumor-specific T cell responses. Combination of radiation with antibodies inhibiting immune checkpoint molecules has shown significant control of the local tumor and of metastatic disease away from the irradiated area. A novel player in innate immune system, the cGAS-STING pathway, up regulates its activity in response to radiation and induces IFN-I-dependent anticancer immunity. The present study focuses on determining the optimal scheme of radiation that elicits cGAS-STING pathway’s activation and IFN-β production. In addition, the HLA-I protein levels of cancer cells is also being examined, over a period of 24hours after radiation. HLA-I is a well characterized indicator of tumor immunogenicity. Results indicate that a repeated dose of 8Gy, given in 3 fractions, amplifies the expression of IFN-I, while single doses of radiation reduce the levels of HLA-I in a dose-dependent manner. Better understanding of the mechanisms by which radiation initiates immune system, is a key point of designing more effective therapeutic strategies.
