The purpose of this research is to study the immune profile of peripheral blood lymphocytes of patients during co-culture of lymphocytes with lung cancer cells. Lymphocytes were isolated from peripheral blood of patients by centrifugation in Ficoll-Hypaque. The separation is based on the fact that the lymphocytes have a lower density than the red blood cells and the granulocytes. We cultured lymphocytes, cancer cell line of lung A549 and co-culture of the above cells. By Western blotting technique we detected the CD4 protein expression (potential regulatory lymphocytes suppress the anti-neoplastic immune response), and PD-1 (immune checkpoint receptor suppressing the action of cytotoxic lymphocytes after PD-L1-binding). With these experiments we found that lymphocyte anti-neoplastic potential is increased after their contact with cancer cells.
