Κυτταρογενετική μελέτη της επίδρασης του φλαβονοειδούς Ναρινγκενίνη και των μεταλλοσυμπλόκων της με Cu (II) και 1,10-φαινανθρολίνη στη δομή των χρωμοσωμάτων (Bachelor thesis)

The present study was carried out in the laboratory of Genetics of the Medical School of the Democritus University of Thrace, under the supervision of Professor Theodoros S. Lialiaris, The purpose of the research is to study the genotoxicity of the complex Cu (II) - Naringenin - 1,10-phenanthroline in cultures of human peripheral blood lymphocytes by studying its effect (and the effect of the constitutant combounds) with the method of sister chromatids exchanges (SCEs). More specifically, the fragility caused by the complex and its constituents is examined in the presence or absence of camptothecin - an anticancer drug - based on the SCEs / Metaphase Index (quantitative index of genotoxicity). In addition, the cytotoxicity of the above was evaluated using two more indicators: the Mitotic Index (a qualitative index of cytotoxicity) and the Proliferation Rate Index (a qualitative index of cytotostaticity). The results showed that at the highest concentrations tested (3 μM & 15 μM) the ΑΔ 211 complex is a highly cytostatic and cytotoxic substance, with these effects even changing in a dose-dependent manner. It also appears that at the concentration of 0.3 μM it showed genotoxic activity when combined with CPT, increasing the fragility caused by the latter. These results indicate that the complex acts in a completely different way compared to its parent flavonoid, as Naringenin appears to have stimulatory role in cell cycle development and gene protection properties at the highest concentration tested (15 μM). In conclusion, the involvement of Naringenin in the ΑΔ 211 complex seems to alter its properties, as it acquires different properties compared to its parent flavonoid.
