Immunophenotypic assessment of measurable residual disease and hematopoietic progenitor cell markers of patients with Acute Myeloid Leukemia in serial timepoints (Bachelor thesis)

Acute Myeloid Leukemia (AML) is a clonal hematological malignancy of the bone marrow caused by genetic alterations in the early stages of hematopoietic stem cell development. The prognosis of the disease depends on the molecular and cytogenetic profile of each patient. However, due to the significant heterogeneity of the disease, some patients relapse because of the presence of leukemic stem cells. These cells are characterized as "Measurable Residual Disease" and are detected using flow cytometry. This method is based on the immunophenotypic characterization of leukemic cells and stem cells. These specific types of cells have unique surface antigens, which are detected by special markers, with the most common being CD133, CD123, and CD33. In this thesis, the immunophenotypic characterization and analysis of leukemic cells and stem cells were performed on bone marrow samples from AML patients who received intensive chemotherapy. The findings of the study revealed that patients who relapsed had increased levels of leukemic stem cells after treatment, indicating that the presence of measurable residual disease could be a strong prognostic factor for the course of the disease. Early detection of these cells flow cytometry can help improve prognosis, allowing for treatment adjustments to prevent relapse.
