heal.abstractΤο ομφαλιοπλακουντικό αίμα αποτελεί πλούσια πηγή αιμοποιητικών στελεχιαίων κυττάρων (HSCs), ενώ ταυτόχρονα περιέχει και άλλους τύπους κυττάρων αίματος. Τα κύτταρα αυτά έχουν την ικανότητα έκκρισης εξωκυτταρικών κυστιδίων, συμπεριλα-μβανομένων των μικροσωματίων, τα οποία σχηματίζονται και εκβλαστάνουν κατά την κυτταρική ενεργοποίηση ή απόπτωση. Ειδικότερα, τα μικροσωμάτια δρουν ως φορείς βιολογικών μορίων, συμμετέχουν στη διακυτταρική επικοινωνία και μπορούν να επηρεάσουν ποικίλες κυτταρικές λειτουργίες. Σκοπός της παρούσας εργασίας ήταν η διερεύνηση της επίδρασης των μικροσωματίων σε καλλιέργεια κυττάρων THP-1 υπό φλεγμονώδεις συνθήκες. Προκειμένου να καθοριστεί η πιθανή ανοσορυθμιστική τους δράση, εφαρμόστηκε in vitro μοντέλο φλεγμονής με τη χρήση λιποπο-λυσακχαρίτη (LPS) προερχόμενου από βακτήρια E. coli. Η αξιολόγηση της επίδρασης των μικροσωματίων παρουσία ή απουσία LPS, πραγματο-ποιήθηκε μέσω ελέγχου της κυτταρικής βιωσιμότητας καθώς και ανάλυσης της γονιδιακής έκφρασης των TLR4, IL-1β, IL-8 και CXCL1. Η προσθήκη LPS (100 ng/mL) και μικροσωματίων δεν επηρέασε τη βιωσιμότητα των κυττάρων THP-1. Δοκιμάστηκαν δύο διαφορετικά πρωτόκολλα απομόνωσης μικροσωματίων. Η έκφραση του TLR4 δεν παρουσίασε στατιστικά σημαντικές διαφορές μεταξύ των πειραματικών ομάδων τόσο στα αδιαφοροποίητα όσο και στα διαφοροποιημένα κύτταρα THP-1. Αντιθέτως, η παρουσία LPS οδήγησε σε αυξημένη έκφραση της κυτοκίνης IL-1β και της χημειοκίνης IL-8, ενώ η επίδραση των μικροσωματίων διαφοροποιήθηκε ανάλογα με το εφαρμοζόμενο πρωτόκολλο απομόνωσης. Συγκεκριμένα, τα μικροσωμάτια που απομονώθηκαν από ολικό πλάσμα αίματος προκάλεσαν αύξηση της έκφρασης των IL-1β και IL-8, ενώ μικροσωμάτια που προέκυψαν μετά από συμπύκνωση του πλάσματος δεν επέφεραν καμία μεταβολή στα κύτταρα. Τέλος, η έκφραση του CXCL1 σημείωσε ήπια αύξηση μόνο παρουσία LPS. Συμπερασματικά, τα αποτελέσματα υποδεικνύουν πως τα μικροσωμάτια δεν επάγουν σημαντικό προφλεγμονώδη φαινότυπο στα μονοκύτταρα και τα μακροφάγα που προέρχονται από τη σειρά THP-1el
heal.abstractUmbilical cord blood (UCB) represents a rich source of hematopoietic stem cells (HSCs), while it also contains other blood cell populations. These cells are capable of releasing extracellular vesicles, including microparticles generated during cellular activation or apoptosis. Microparticles act as carriers of bioactive molecules, participate in intercellular communication, and may influence multiple cellular processes. The aim of the present study was to investigate the effect of microparticles on THP-1 cell cultures under inflammatory conditions. To determine their potential immunomodulatory properties, an in vitro inflammation model was applied using lipopolysaccharide (LPS) derived from E. coli. The impact of microparticles’, in the presence or absence of LPS, was evaluated by assessing cell viability and analyzing the gene expression levels of TLR4, IL-1β, IL-8 and CXCL1. The the addition of LPS (100 ng/mL) or microparticles did not affect THP-1 cells viability. Two different microparticle isolation protocols were evaluated. TLR4 expression did not demonstrate statistically significant differences among the experimental groups in either undifferentiated or differentiated THP-1 cells. In contrast, LPS stimulation increased the expression of the cytokine IL-1β and the chemokine IL 8, while the effect of microparticles varied depending on the isolation protocol employed. Specifically, microparticles isolated from whole blood plasma induced an increase in IL-1β and IL-8 expression, whereas microparticles obtained after plasma concentration did not alter gene expression levels. CXCL1 expression showed only a mild increase in response to LPS. Overall, the findings suggest that microparticles do not promote a pro-inflammatory phenotype in THP-1 derived monocytes and macrophages.en
